Optical Thickness Measurement System

Optical thickness measurement system (OTMS) set up

  1. The frame is built using MakerBeam framing system.

  2. Lens: Fujifilm lens HF12.5HA-1B
  3. Camera: Firefly fmvu-13s2c-cs
  4. Ruler/gage pin: Stainless steel precision ruler
  5. Background: Blue

OTMS image acquisition

  1. Place the sample close to the edge of the platform.
  2. Use flycapture software to acquire tissue and ruler image.
  3. From the file menu capture video.
  4. Choose the folder where image is going to be saved, Frames=1, file type= .jpg.
  5. Refer to Specifications/DataManagement for naming conventions.

Using the GUI

  1. Run the script from the command prompt.

  2. Choose browse from the GUI to find the desired .jpg file and open the image.
  3. First, choose the maximum measurement along the ruler and the zero point at the bottom of the ruler. Enter the measurement (mm) in the box next to the calibrate button.
  4. Hit "Calibrate". This should display the number of pixels in the window.
    • For all pick point sets, try to choose near the same X axis value to create vertical lines.
  5. Next, choose "Pick", then choose the two points to create a line connecting the upper and lower boundaries of the sample, hit "OK". Proceed to pick at least ten sets of points.
  6. Choose "compute" to find the average, providing the sample thickness.
